چکیده

Incubation o f deproteinized larval cuticle (chitin flakes) with purified arylphorin (calliphorin) or larval haemolymph o f Calliphora vicina resulted in the formation o f a chitin-protein complex. Enzymatic oxidation o f N-ß-alanyldopamine (N D A B ) in the presence o f chitin flakes or the chitin-protein complex, resulted in various degrees o f cross-linking o f N BA D quinone formed with chitin. This study confirms the involvement o f arylphorin in the process o f quinone tanning o f insect cuticle.

Model reactions for insect cuticle sclerotization: participation of amino groups in the cross-linking of Manduca sexta cuticle protein MsCP36.

Current theories of sclerotization center on protein cross-linking and dehydration as major factors in the hardening and stability of the insect cuticle. Several studies have reported the identification of catechol-amino acid adducts from sclerotizing cuticle involving histidine, lysine, and tyrosine, though there have been no reports of a catechol linked between two amino acid residues. Previo...

Great Britain 3 - 0 - Hydrosulphato 4 hydroxyphenethylamine ( Dopanine 3 - 0 - Sulphate ) , a Metabolite Involved in the Sclerotization of Insect Cuticle

Dopamine 3-0-sulphate (3-0-hydrosulphato-4-hydroxyphenethylamine) was isolated from newly ecdysed cockroaches, Periplaneta americana (L.), and its structure established by chemical and physical techniques and by synthesis. Relatively high concentrations (about 1 pmol/g wet wt.) of dopamine 3-0-sulphate exist in the newly ecdysed insect, and these concentrations decrease sharply as sclerotizatio...

Identification of the Gene Encoding Bursicon, an Insect Neuropeptide Responsible for Cuticle Sclerotization and Wing Spreading

To accommodate growth, insects must periodically replace their exoskeletons. After shedding the old cuticle, the new soft cuticle must sclerotize. Sclerotization has long been known to be controlled by the neuropeptide hormone bursicon, but its large size of 30 kDa has frustrated attempts to determine its sequence and structure. Using partial sequences obtained from purified cockroach bursicon,...

Model reactions for insect cuticle sclerotization: cross-linking of recombinant cuticular proteins upon their laccase-catalyzed oxidative conjugation with catechols.

The quinone-tanning hypothesis for insect cuticle sclerotization proposes that N-acylcatecholamines are oxidized by a phenoloxidase to quinones and quinone methides, which serve as electrophilic cross-linking agents to form covalent cross-links between cuticular proteins. We investigated model reactions for protein cross-linking that occurs during insect cuticle sclerotization using recombinant...

Incorporation of Radiolabeled Tyrosine, N-Acetyldopamine, N-/J-Alanyldopamine, and the Arylphorin Manducin into the Sclerotized Cuticle of Tobacco Hornworm (Manduca sexta) Pupae

Radiolabeled manducin, which is the arylphorin (i.e. tyrosine-rich larval serum protein) o f the tobacco hornworm Manduca sexta was prepared by in vivo biosynthesis from [U -l4C]-L-tyrosine. In order to see whether manducin is incorporated into the cuticle o f developing pupae and participates in cuticle sclerotization, the radiolabeled protein was injected into late last larval instar larvae. ...
